Evaluation of the automated system Vitek2 for identification and antimicrobial susceptibility testing of Brazilian Gram-positive cocci strains BJID
d'Azevedo,Pedro Alves; Siquiera,Itacy; Gugel,Juliana; Antunes,Ana Lúcia Souza; Secchi,Carina; Pasternak,Jacyr; Martino,Marinês Dalla Vale.
Automated instruments offer many advantages for clinical laboratories. Nevertheless, they can have problems identifying and determining susceptibilities of some pathogens. Vitek® 2 (bioMérieux) is an automated system that was recently introduced to Brazil. We evaluated the performance of this equipment for Brazilian isolates that had been characterized using reference identification and antimicrobial susceptibility testing methods. Ninety-nine strains of Gram-positive cocci from a local reference center collection were analyzed, consisting of 50 coagulasenegative Staphylococcus (CoNS) and 49 Enterococcus and related species. Vitek® 2 correctly identified 79.8% (79/99) of the isolates. Oxacillin resistance was detected in 76% (19/25) of resistant S....

Identificação dos insetos infestantes de alimentos através da micromorfologia de seus fragmentos Zoologia
Vargas,Carlos Henrry Bellot; Almeida,Armando Antunes de.
The identification of insect fragments showed that the infestation was done, mainly, by Coleoptera of the genus Sitophilus Sch›nherr, 1838 (Curculionidae) and Rhizoperthadominica (Fabricius, 1792) (Bostrychidae), that are caracteristic of internai infestation, and in lesser percentage by insects charac-teristic of externai infestation, as Crylolestes spp. (Cucujidae), Tribolium spp. (Tene-brionidae), Oryzaephilussurinamensis(Linnacus, 1758) (Cucujidae) and Lasioderma serricorne (Fabricius, 1792) (Anobiidae). From the total of recovered and identified insect fragments, 59% were fragments of elytra, 23% of mandibles, and in minor percentage by tarsal segments, claws, tibial spurs, legs, tarsus, eyes, antennae, exuviae and larvae urogomphi.

Identification and adhesion profile of Lactobacillus spp. strains isolated from poultry BJM
Rocha,Ticiana Silva; Baptista,Ana Angelita Sampaio; Donato,Tais Cremasco; Milbradt,Elisane Lenita; Okamoto,Adriano Sakai; Andreatti Filho,Raphael Lucio.
In the aviculture industry, the use of Lactobacillus spp. as a probiotic has been shown to be frequent and satisfactory, both in improving bird production indexes and in protecting intestine against colonization by pathogenic bacteria. Adhesion is an important characteristic in selecting Lactobacillus probiotic strains since it impedes its immediate elimination to enable its beneficial action in the host. This study aimed to isolate, identify and characterize the in vitro and in vivo adhesion of Lactobacillus strains isolated from birds. The Lactobacillus spp. was identified by PCR and sequencing and the strains and its adhesion evaluated in vitro via BMM cell matrix and in vivo by inoculation in one-day-old birds. Duodenum, jejunum, ileum and cecum were...

Identification and in vitro production of Lactobacillus antagonists from women with or without bacterial vaginosis BJMBR
Branco,K.M.G.R.; Nardi,R.M.D.; Moreira,J.L.S.; Nunes,A.C.; Farias,L.M.; Nicoli,J.R.; Carvalho,M.A.R..
Lactobacilli isolated from the vaginal tract of women with and without bacterial vaginosis (BV) were identified and characterized for the production of antagonists. Bacterial samples were isolated from healthy women (N = 16), from patients with clinical complaints but without BV (N = 30), and from patients with BV (N = 32). Identification was performed using amplified ribosomal DNA restriction analysis. Production of antagonistic compounds was evaluated by the double-layer diffusion technique using Gram-positive (N = 9) and Gram-negative bacteria (N = 6) as well as yeast (N = 5) as indicator strains. Of a total of 147 isolates, 133 were identified as pertaining to the genus Lactobacillus. Lactobacillus crispatus was the species most frequently recovered,...

Identification of a novel gene encoding a secreted antigen 1 of Babesia gibsoni and evaluation of its use in serodiagnosis OAK
Jia, Honglin; Zhou, Jinlin; Ikadai, Hiromi; Matsuu, Aya; Suzuki, Hiroshi; Fujisaki, Kozo; Xuan, Xuenan; 鈴木, 宏志; 五十嵐, 郁男; 玄, 学南.
Serum from a dog immunized with blood plasma from a B. gibsoni-infected dog, putatively containing secreted antigens, was used to screen a cDNA expression library. A novel gene encoding BgSA1 was identified from the isolated clones. The serum raised in mice immunized with the recombinant BgSA1 expressed in Escherichia coli could recognize a native parasite protein with a molecular mass of 59 kDa. Comparing with the previously established ELISA with recombinant P50 as antigen, the ELISA with recombinant BgSA1 as the antigen was more sensitive when they were used to detect field samples. Moreover, a sandwich ELISA with anti-BgSA1 antibodies could detect the circulating BgSA1 in a serial blood plasma from a dog experimentally infected with B. gibsoni. These...

Identification of Candida spp. by phenotypic tests and PCR BJM
Marinho,Sandra Aparecida; Teixeira,Alice Becker; Santos,Otávio Silveira; Cazanova,Ricardo Flores; Ferreira,Carlos Alexandre Sanchez; Cherubini,Karen; Oliveira,Sílvia Dias de.
The correct identification of Candida species is of great importance, as it presents prognostic and therapeutical significance, allowing an early and appropriate antifungical therapy. The purpose of this study was to identify isolates of Candida spp. from oral mucosa of 38 patients with oral candidosis evaluated in 2004 by phenotypic methods and PCR, discriminating C. albicans from the other Candida species. The tests used for phenotypic analysis were germ-tube and chlamydoconidia production, culture in CHROMAgarTM Candida, carbohydrate assimilation test, growth at 45ºC and culture in Tween 80 agar. Genotypic confirmation was performed by PCR. Phenotypic tests showed that 63.2% strains formed germ-tubes, 73.7% produced chlamydoconidia, and 63.2% showed...

IDENTIFICATION OF ECONOMIES OF SCOPE IN A STOCHASTIC PRODUCTION ENVIRONMENT AgEcon
Curtis, Rita E.; Sarmiento, Camilo.
This paper extends the definition of economies of scope to multioutput firms that face an uncertain production environment. Identification of economies of scope in this environment, however, requires separability assumptions on the technology. These identification restrictions are demonstrated in the paper, and for each identification restriction the definition of economies of scope is generalized to the case of uncertain production and risk aversion.

Identification of fungemia agents using the polymerase chain reaction and restriction fragment length polymorphism analysis BJMBR
Santos,M.S.; Souza,E.S.; Junior,R.M.S.; Talhari,S.; Souza,J.V.B..
Prompt and specific identification of fungemia agents is important in order to define clinical treatment. However, in most cases conventional culture identification can be considered to be time-consuming and not without errors. The aim of the present study was to identify the following fungemia agents: Candida albicans, Candida parapsilosis, Candida tropicalis, Candida glabrata, Cryptococcus neoformans, Cryptococcus gattii, and Histoplasma capsulatum using the polymerase chain reaction and restriction fragment length polymorphism analysis (PCR/RFLP). More specifically: a) to evaluate 3 different amplification regions, b) to investigate 3 different restriction enzymes, and c) to use the best PCR/RFLP procedure to indentify 60 fungemia agents from a culture...

Identification of non-tuberculous mycobacteria from the Central Public Health Laboratory from Mato Grosso do Sul and analysis of clinical relevance BJM
Moraes,Paulo Ricardo de Souza; Chimara,Erica; Telles,Maria Alice da Silva; Ueki,Suely Yoko Misuka; Cunha,Eunice Atsuko Totumi; Honer,Michael Robin; Leão,Sylvia Cardoso.
Non-tuberculous mycobacteria isolated at the Central Public Health Laboratory from Mato Grosso do Sul in 2003 and 2004 were identified by conventional phenotypic methods (TI) and by PCR-Restriction Enzyme Analysis (PRA) using the hsp65 gene as target (PRA-hsp65). With 15 of the 32 analysed isolates, results of both methods were concordant, being 8 Mycobacterium avium, 3 M. fortutium, 1 M. kansasii, 1 M. flavescens, 1 M. peregrinum and 1 Nocardia brasiliensis. TI of 12 isolates was inconclusive. Novel PRA-hsp65 patterns were observed with 11 isolates. Medical data were evaluated for inference of clinical relevance of these isolates.

Identification of pathogenic and nonpathogenic Leptospira species of Brazilian isolates by Matrix Assisted Laser Desorption/Ionization and Time Flight mass spectrometry BJM
Karcher,Daniel; Grenfell,Rafaella C.; Moreno,Andrea Micke; Moreno,Luisa Zanolli; Vasconcellos,Silvio Arruda; Heinemann,Marcos B.; Almeida Junior,Joao N. de; Juliano,Luiz; Juliano,Maria A..
ABSTRACT Matrix Assisted Laser Desorption/Ionization and Time of Flight mass spectrometry (MALDI-TOF MS) is a powerful tool for the identification of bacteria through the detection and analysis of their proteins or fragments derived from ribosomes. Slight sequence variations in conserved ribosomal proteins distinguish microorganisms at the subspecies and strain levels. Characterization of Leptospira spp. by 16S RNA sequencing is costly and time-consuming, and recent studies have shown that closely related species (e.g., Leptospira interrogans and Leptospira kirschneri) may not be discriminated using this technology. Herein, we report an in-house Leptospira reference spectra database using Leptospira reference strains that were validated with a collection...

Identification of Supply Models of Retailer and Manufacturer Oligopoly Pricing AgEcon
Villas-Boas, Sofia Berto; Hellerstein, Rebecca.
This note outlines conditions under which we can identify a vertical supply model of multiple retailers' and manufacturers' oligopoly-pricing behavior. This is an important question particularly when the researcher believes, contrary to the traditional assumption followed in the empirical literature, that retailers may not be neutral pass-through intermediaries. We show that a data-set of an industry's product prices, quantities, and input prices over time is sufficient to identify the vertical model of retailers' and manufacturers' oligopoly-pricing behavior given nonlinear demand, for homogeneous-products industries, and given multi-product firms, for differentiated-products industries.
